Immunoprecipitation: BRD4 Antibody [NBP1-18874] - Detection of human BRD4 by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-BRD4 antibody NBP1-18874 used for IP at 3 ug per reaction. BRD4 was also immunoprecipitated by a different rabbit anti-BRD4 antibody and rabbit anti-BRD4 recombinant monoclonal antibody [BL-149-2H5] (NBP2-76393). Q: Why is BRD4 detected at around 200kd when it is a 152kd protein? Additionally, I noticed another company's antibody detects a protein of around 85kd.
A: Our antibody is detecting the full length BRD4 protein 1362 amino acids in length, which corresponds to approximately 200kDa. This is also referred to as the long isoform of BRD4. The 85kDa band that you mention being detected by another antibody would most likely correspond to the short isoform of BRD4 which is the alternative splice variant. The size of the short isoform is 722 amino acids which corresponds to approximately 83.45kDa.
