This antibody was raised by immunizing mice with a synthetic phosphopeptide (CEAPERA(pS)(pS)VYTR(pS)TGEQI(pS)VGL) corresponding to the C-terminal region of human CCR5.

The E11/19 monoclonal antibody binds to phosphoserine 349 corresponding to 22 c-terminal amino acid residue of human CCR5. CCR5 is a receptor for a number of inflammatory CC-chemokines including CCL3/MIP-1-alpha, CCL4/MIP-1-beta and RANTES.

Western Blot: CCR5 [p Ser349] Antibody (E11/19) - Chimeric [NBP3-11995] - Molt4 cell lysate (35ug protein in RIPA buffer) was resolved on a SDS PAGE gel and blots were probed with the chimeric rabbit version of E11/19 (NBP3-11995) at 0.1 ug/ml before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.
Immunocytochemistry/Immunofluorescence: CCR5 [p Ser349] Antibody (E11/19) - Chimeric [NBP3-11995] - Immunofluorescence analysis of paraformaldehyde fixed U937 cells on Shi-fix(TM) coverslips stained with the chimeric rabbit IgG version of E11/19 (NBP3-11995) at 10 ug/ml for 1h followed by Alexa Fluor(R) 488 secondary antibody (2 ug/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP3-11995, DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody followed by staining with Alexa Fluor(R) 488 secondary antibody.

Flow Cytometry: CCR5 [p Ser349] Antibody (E11/19) - Chimeric - Azide and BSA Free [NBP3-11995] - U937 cells were fixed using 2% PFA and stained with anti-unknown specificity antibody (isotype control, black line) or NBP3-11995 (blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.Applications for CCR5 [p Ser349] Antibody (E11/19) - Chimeric - Azide and BSA Free

Background: CCR5
CCR5 is a major coreceptor for human HIV infection and is predominantly expressed on T cells, macrophages, dendritic cells and microglia. It is likely that CCR5 plays a role in inflammatory responses to infection, though its exact role in normal immune function is unclear.
CCR5 antibodies are useful for HIV infection studies and research on immune responses.
