Detection of Human CD133 by Simple WesternTM.Dilution: 4-20 µg/mL
Left: Simple Western lane view shows lysates of HT‑29 human colon adenocarcinoma cell line, loaded at 0.1 mg/ml. A specific band was detected for CD133 at approximately 166 kDa (as indicated) using both 4 µg/ml and 20 µg/ml of Mouse Anti-Human CD133 Monoclonal Antibody (Catalog # NBP3-32124) followed by HRP-conjugated Goat Anti-Mouse Secondary Antibody (Catalog # 042-205). This experiment was conducted under reducing conditions and using the 12-230kDa separation system. Western Blot: CD133 Antibody (A8C3-R) [NBP3-32124] - Western blot analysis of CD133 on different lysates with Mouse anti-CD133 antibody (NBP3-32124) at 1/5,000 dilution.Lane 1: HT-29 cell lysate

Exposure time: 2 minutes;
10% SDS-PAGE gel.
Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32124) at 1/5,000 dilution was used in 5% NFDM/TBST at room temperature for 2 hours. Goat Anti-Mouse IgG - HRP Secondary Antibody at 1:100,000 dilution was used for 1 hour at room temperature.

Immunohistochemistry: CD133 Antibody (A8C3-R) [NBP3-32124] - Immunohistochemical analysis of paraffin-embedded human colon carcinoma tissue with Mouse anti-CD133 antibody (NBP3-32124) at 1/1,000 dilution.The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (NBP3-32124) at 1/1,000 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
