Application Notes
The 5C3 antibody has been tested by flow cytometric analysis of human peripheral blood leukocytes. This can be used at less than or equal to 1 ug per test. Cell number should be determined empirically but can range from 10^5to 10^8cells/test. This antibody may also be used for immunohistochemical staining of frozen tissue sections. 5C3 has also been reported in in vitro functional studies.

Background: CD40/TNFRSF5

The 5C3 monoclonal antibody reacts with human CD40, a 45-50 kDa type I transmembrane glycoprotein. CD40 is a member of the TNFR family and is expressed by B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells. CD40 regulates B cell development and maturation by inducing Ig isotype-switching and in combination with other signals such as IL-4, protects B cells from surface Ig-induced apoptosis and promotes proliferation. Interaction of CD40 with CD154 (gp39), its ligand on T cells, is important in T-B cell crosstalk and plays a role in costimulation and immune regulation. 5C3 is reported to be used for activation of B cells in in vitro functional assays.

  • Q: I wanna know does CD40 antibody for IHC-P comes with positive control slide?

    A: Our CD40 antibodies do not come with positive control slides however you can use any type of primary carcinoma cell as a positive control. I recommend breast carcinoma as this is what we have tested our CD40 antibodies on.

  • Q: We are studying CD40L in our system. It has been known that both CD40 and CD11b are receptors for CD40L. We need CD40 antibody to neutralize CD40 binding to CD40L, then we may prove what is function for CD11b binding to CD40L. Do you have any CD40 Ab which has block function only between CD40 and CD40L, but not between CD11b and CD40L?

    A: Unfortunately, none of our CD40 antibodies have been specifically tested for this application.
