Western Blot: CD43/Sialophorin Antibody (W3/13) [NB100-64992] - Western blot shows lysates of rat lymph node, rat spleen tissue, and rat thymus tissue. PVDF membrane was probed with 0.5 ug/mL of Mouse Anti-Rat CD43 Monoclonal Antibody (Catalog # NB100-64992) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for CD43 at approximately 90 kDa (as indicated). This experiment was conducted under reducing conditions.

Immunohistochemistry-Paraffin: CD43/Sialophorin Antibody (W3/13) [NB100-64992] - CD43 was detected in immersion fixed paraffin-embedded sections of rat thymus using Mouse Anti-Rat CD43 Monoclonal Antibody (Catalog # NB100-64992) at 5 ug/mL for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC001). Before incubation with the primary antibody, tissue was incubated with VisUBlock™ Mouse on Mouse Blocking Reagent (Catalog # VB001) and then subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to lymphocytes.

Background: CD43

CD43 is one of the major glycoproteins of thymocytes and T lymphocytes, it plays a role in the physicochemical properties of the T cell surface and in lectin binding. CD43 presents carbohydrate ligands to selectins. It has an extended rodlike structure that could protrude above the glycocalyx of the cell and allow multiple glycan chains to be accessible for binding. The antigen is a counter receptor for SN/Siglec1. During T cell activation CD43 is actively removed from the T cell antigen presenting cell contact site suggesting a negative regulatory role in adaptive immune response. CD43 is expressed by the majority of leucocytes, with the exception of most B cells. However CD43 expression is seen in some cases of B cell lymphocytic lymphoma and centrocytic lymphoma. CD43 cell surface antigen, also known as leukosialin and sialophorin. CD43 is a 95kD heavily glycosylated glycoprotein expressed by all leucocytes with the exception of B lymphocytes. This product is routinely tested in flow cytometry on rat splenocytes.
