Western Blot: Cytochrome P450 17A1 Antibody (JB93-32) [NBP2-75461] - Analysis of Cytochrome P450 17A1 on rat testis tissue lysates with Rabbit anti-Cytochrome P450 17A1 antibody at 1/500 dilution. Lysates/proteins at 20 ug/Lane. Predicted band size: 57 kDa Observed band size: 55 kDa Exposure time: 2 minutes; 10% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody at 1/500 dilution was used in 5% NFDM/TBST at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:300,000 dilution was used for 1 hour at room temperature.

Background: Cytochrome P450 17A1

The CYP17A1 gene encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. The CYP17A1 protein localizes to the endoplasmic reticulum. CYP17A1 has both 17alpha-hydroxylase and 17,20-lyase activities and is a key enzyme in the steroidogenic pathway that produces progestins, mineralocorticoids, glucocorticoids, androgens, and estrogens. Mutations in the CYP17A1 gene are associated with isolated steroid-17 alpha-hydroxylase deficiency, 17-alpha-hydroxylase/17,20-lyase deficiency, pseudohermaphroditism, and adrenal hyperplasia.
