Background: Cytokeratin 1

Cytokeratin 1 is a heterotetramer of two type I as well as two type II keratins localized in cell membrane generally associated with keratin-10 (KRT10), and it has been suggested to regulate the activity of kinases such as PKC/SRC via binding to integrin beta-1 (ITB1) and the receptor of activated protein kinase C (RACK1/GNB2L1). Cytokeratin 1 and its heterodimer partner KRT10 are the major constituents of 4 intermediate filament cytoskeleton in suprabasal epidermis/keratinocytes, and their upregulation precedes expression of filaggrin and cornified envelope proteins. Covalent crosslinking of cytokeratin 1 but not of KRT10 to these proteins, in addition to a complex series of lipids, ultimately forms the cornified/lipid envelope, that constitutes the epidermal barrier together with Langerhans cells. Defects in cytokeratin 1 have been linked to epidermolytic hyperkeratosis (EHK), ichthyosis hystrix Curth-Macklin type (IHCM), palmoplantar keratoderma non-epidermolytic (NEPPK), ichthyosis annular epidermolytic (AEI), palmoplantar keratoderma striate type 3 (SPPK3).
