Western Blot Shows EPHX2 Specificity Using Knockout Cell Line.

Western blot shows lysates of K562 human chronic myelogenous leukemia parental cell line and EPHX2 knockout K562 cell line (KO). Nitrocellulose membrane was probed with EPHX2 Antibody (OTI1A6) (Catalog # NBP2-02948) followed by HRP-conjugated secondary antibody. A specific band was detected for EPHX2 at approximately 62.6 kDa (as indicated) in the parental K562 cell line, but is not detectable in knockout K562 cell line. Primary antibody dilution used: 1/500. The Ponceau stained transfer of the blot is shown. This experiment was conducted under reducing conditions. Background: Epoxide Hydrolase 2/EPHX2

This gene encodes a member of the epoxide hydrolase family. The protein, found in both the cytosol and peroxisomes, binds to specific epoxides and converts them to the corresponding dihydrodiols. Mutations in this gene have been associated with familial hypercholesterolemia. Alternate transcriptional splice variants of this gene have been observed but have not been thoroughly characterized.
