Histone H2B synthetic peptide (coupled to carrier protein) corresponds to amino acids in the C-terminus of human histone H2B. This sequence is well conserved in rat, mouse, fish, and drosophila histone H2B, and has low homology to other histone family members. [UniProt# P33778]

Specificity

The antibody detects a 15 kDa protein corresponding to the molecular mass of Histone H2B type 1B on SDS-PAGE immunoblots of human A431, Jurkat, HeLa, mouse C2C12, rat PC12, and A7r5 cells. The antibody also shows nuclear labeling in immunocytochemistry of methanol/acetone fixed cells.

Immunocytochemistry/Immunofluorescence: Histone H2B type 1B Antibody [NBP3-23376] - Immunocytochemical labeling of Histone H2B type 1B in methanol and acetone fixed rat A7r5 cells. The cells were labeled with rabbit polyclonal Histone H2B type 1B (C-terminus) antibody (NBP3-23376), then the antibody was detected using appropriate secondary antibody conjugated to DyLight(R) 594.

The blots were probed with anti-Histone H2B type 1B (C-terminus) (lanes 1 & 2), anti-Histone H2B type 1B (Ser-36) (lanes 3 & 4), and anti-Histone H2B type 1B (a.a. 33-47) (lanes 5 & 6).

The HIST1H2BB gene encodes a 126 amino acid long, 13 kDA histone H2B type 1-B that is critical in the nucleosome by functioning in transcription regulation, DNA replication and repair, as well as chromosomal stability. It participates in histone modification, apoptosis and authophagy, protein stability, RSF complex binding to the centromer, and signal transduction Activin A signaling regulation. HIST1H2BB is known to interact with genes EP300, ERCC6, DYRK1A, NFKB2, and TNFRSF1A. It has been researched regarding its role to lupus, immunodeficiency, cockayne syndrome, and hemochromatosis.
