Gab2 (GRB2-associated binding protein 2) is a 95 kDa member of the Gab family of proteins. It is largely expressed in T and B cells, and serves as an adaptor molecule for the transmission of signals from cytokine and growth factor receptors. Bcr-Abl signals through Gab2, and Gab2 activates ERK. Human Gab2 is 676 amino acids (aa) in length, and contains one pleckstrin homology domain (aa 6‑117) and one methyl binding domain sequence (aa 454‑525). Multiple phosphorylation sites exist, with S159 being an important regulatory site. One alternate start site is reported at M39. Over aa 82‑292, human Gab2 is 93% and 98% aa identical to mouse and canine Gab2, respectively.

Detection of Human Gab2 by Western Blot.
Western blot shows lysates of MO7e human megakaryocytic leukemic cell line, U937 human histiocytic lymphoma cell line, and A549 human lung carcinoma cell line. PVDF membrane was probed with 0.2 µg/mL of Human Gab2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4877) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for Gab2 at approximately 95 kDa (as indicated).
