|Detection of Mouse IkB-beta by Western Blot. Western blot shows lysates of NIH‑3T3 mouse embryonic fibroblast cell line, BaF3 mouse pro‑B cell line, M1 mouse myeloid leukemia cell line. PVDF membrane was probed with 1 µg/mL of Human/Mouse IkB-beta Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5225) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). A specific band was detected for IkB-beta at approximately 50 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.|
I kappa B-beta (I kappa B-beta; also NF-kappa-BIB) is a 46 kDa member of the I kappa B-beta inhibitor family of proteins. It is highly expressed in testicular spermatids, and serves to block NF-kappa B activity. This is accomplished by I kappa B-beta binding to either an NF-kappa B dimer, or a kappa B-Ras:NF-kappa B dimer complex. Mouse I kappa B-beta is 359 amino acids (aa) in length it contains a signal response region (aa 1‑50), six ankyrin repeats (aa 54‑308) and a PEST domain whose phosphorylation at Ser312 and Ser314 blocks NF‑ kappa B activation. Phosphorylation at Ser19 and Ser23 promotes I kappa B‑ beta ubiquitination and degradation. Full‑length mouse I kappa B‑ beta shares 94% and 84% aa identity with rat and human I kappa B-beta, respectively.
