|Detection of Human, Mouse, and Rat Properdin by Western Blot. Western blot shows lysates of human serum, mouse serum, and rat serum. PVDF membrane was probed with 1 µg/mL of Sheep Anti-Human/Mouse/Rat Properdin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8216) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for Properdin at approximately 53-55 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.|
Human Properdin, also known as Complement factor P (CFP), is a 53 kDa secreted glycoprotein that exists in plasma as dimers, trimers or tetramers. It a positive regulator of the alternate pathway of complement. Properdin binds to and stabilizes the C3- and C5-convertase enzyme complexes and is thought to connect humoral and cellular immunity. Properdin released from activated neutrophils may play a major role in pattern recognition, whereas plasma properdin may primarily stabilize convertase. Properdin deficiency results in higher susceptibility to bacterial infections. Full-length human Properdin shares 77 % sequence identity with mouse Properdin.
