Activity

Recombinant Mouse Wnt-3a induces beta -catenin responsive Topflash activation in HEK293T cells. The specific Wnt pathway inhibitor, endo-IWR 1, inhibits Topflash activity from a 500 ng/mL dose of Recombinant Mouse Wnt-3a with a typical ED50 < 0.2 μM. The less active diasteriomer exo-IWR 1 Wnt inhibitor shows minor inhibitory activity in this assay.

Recombinant Mouse Wnt-3a induces alkaline phosphatase production in the MC3T3-E1 mouse pre-osteoblast cell line. The ED50 for this effect is ≤ 5 ng/mL. The specific Wnt pathway inhibitor, endo-IWR 1, inhibits alkaline phosphatase production induced from a 10 ng/mL dose of Recombinant Mouse Wnt-3a with a typical ED50 < 0.8 μM. The less active diasteriomer exo-IWR 1 Wnt inhibitor shows a > 5 μM ED50 with a Recombinant Mouse Wnt-3a dose of 10 ng/mL.

Background: Wnt-3a

Wnt-3a is a secreted glycoprotein that is necessary for proper development of the hippocampus, anterior-posterior patterning, somite development, and tailbud formation (1-4). Wnt-3a also promotes self-renewal of hematopoietic stem cells (5,6). Wnt-3a, one of 19 vertebrate members of the Wnt family, binds to Frizzled receptor proteins along with LRP5/6 transmembrane proteins causing an intracellular cascade ultimately resulting in increased intracellular beta -catenin levels (7). As intracellular beta -catenin levels rise, beta -catenin binds to TCF/LEF transcription factors leading to Wnt/ beta -catenin-responsive gene activation (8). The intracellular protein Axin is a critical scaffold protein necessary for assembly of the beta -catenin destruction complex that consists of APC, GSK-3 beta, and beta -catenin. Once bound to this complex, beta -catenin is phosphorylated and targeted for degradation (9). Endo-IWR 1 is a small molecule inhibitor of Axin turnover resulting in stabilization of the beta -catenin destruction complex and suppression of Wnt signal transduction. The diastereomeric form of IWR 1, exo-IWR 1, exhibits decreased Axin stabilizing activity compared to endo-IWR 1 and can be used as a specificity control (10).
