NTH1 Antibody (22H10)

Immunohistochemistry: NTH1 Antibody (22H10) [NBP3-26456] -

Immunohistochemistry: NTH1 Antibody (22H10) [NBP3-26456] - Image of NTH1 Antibody (22H10) diluted at 1:100 and staining in paraffin-embedded human testis tissue performed. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB.
NTH1 Antibody (22H10)

Western Blot: NTH1 Antibody (22H10) [NBP3-26456] -

Western Blot: NTH1 Antibody (22H10) [NBP3-26456] - Positive Western Blot detected in: HepG2 whole cell lysate, HEK293 whole cell lysate, PC-3 whole cell lysate.
All lanes: NTH1 Antibody at 1: 1000
Secondary: Goat polyclonal to rabbit IgG at 1/50000 dilution.
Predicted band size: 35, 34, 33 kDa
Observed band size: 35 kDa

Immunocytochemistry/Immunofluorescence: NTH1 Antibody (22H10) [NBP3-26456] -

Immunocytochemistry/Immunofluorescence: NTH1 Antibody (22H10) [NBP3-26456] - Staining of PC-3 cell with NTH1 Antibody (22H10) at 1:50, counter-stained with DAPI. The cells were fixed in 4% formaldehyde and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4C. The secondary antibody was Alexa Fluor 604-conjugated Goat Anti-Rabbit IgG (H+L).

Flow Cytometry: NTH1 Antibody (22H10) [NBP3-26456] -

Flow Cytometry: NTH1 Antibody (22H10) [NBP3-26456] - Overlay Peak curve showing HepG2 cells stained with NTH1 Antibody (22H10) (red line) at 1:100. The cells were fixed in 4% formaldehyde and permeated by 0.2% Triton X-100. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*10^6 cells) for 45min at 4C. The secondary antibody used was FITC-conjugated Goat Anti-rabbit IgG (H+L) at 1:200 dilution for 35min at 4C. Control antibody (green line) was rabbit IgG (1ug/1*10^6 cells) used under the same conditions. Acquisition of >10,000 events was performed.

Background: NTH1

Oxygen radicals damage chromosonal DNA causing cell death and inducing mutations. Although a major focus of oxidatively damaged DNA has centered on the repair of 8-oxo-G, a number of other damaged DNA sites are created by free-radical attack on DNA. The human NTH1 repair protein is one enzyme that has been shown to act on a large number of these other oxidatively damaged DNA sites. A homologue of E. coli Endonuclease III, NTH is a DNA glycosylase with apurinic/apyrimidinic lyase activity. The NTH protein has broad substrate specificity, including numerous ring saturation and fragmentation products of pyrimidines. Research indicates NTH plays a crucial role in removal of oxidative base lesions in mitochondrial DNA.
