Western Blot: PAK1/2/3 [p Ser144, p Ser141, p Ser139] Antibody (3H12) [NBP3-15222] - Positive WB detected in A549 whole cell lysate (treated with EGF or not). All lanes: PAK1/2/3 antibody at 1.2ug/ml. Secondary: Goat polyclonal to rabbit IgG at 1/50000 dilution. Predicted band size: 65 KDa. Observed band size: 65 KDa.
Immunohistochemistry: PAK1/2/3 [p Ser144, p Ser141, p Ser139] Antibody (3H12) [NBP3-15222] - IHC image of PAK1/2/3 antibody diluted at 1:100 and staining in paraffin-embedded rat brain tissue. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

Background: PAK1/2/3

In mammals, there are several identified isoforms of p21-activated Protein Kinases or PAKs: alpha-PAK (also known as PAK-1) and beta-PAK (also known as PAK-3) are mostly brain-specific, while gamma-PAK (also known as PAK-2) is expressed ubiquitously. Mutations of the gene coding for PAK-3 are associated with X-linked mental retardation and recent work indicates that PAK-3 is a key regulator of synapse formation and plasticity in the hippocampus. PAK-3 is thought to play a key role in regulation of cell shape and motility as well as cell death. Autophosphorylation of Thr402 in the protein has been found to be essential for activation of PAK.
