PIEZO1 in MCF7 Human Cell Line.

PIEZO1 was detected in immersion fixed MCF7 human breast cancer cell line using Rabbit anti-PIEZO1 Affinity Purified Polyclonal Antibody conjugated to DyLight 650 (Catalog # NBP1-78446C) (light blue) at 10 µg/mL overnight at 4C. Cells were stained counterstained with DAPI (blue). Cells were imaged using a 100X objective and digitally deconvolved.

Background: PIEZO1

Piezo1 (Mib, FAM38A) is a mechanosensitive ion channel which responds to mechanically activated currents in numerous cells, including the brain, bladder, colon, kidney, lung, and skin. However, mRNA expression of Piezo1 is weak in the DRG, unlike Piezo2. The expression of Piezo1 is induced in Alzheimer's associated senile plaques (PMID 16854388), as well as integrin activation through R-Ras signaling (PMID 20016066). R-Ras is recruited to the endoplasmic reticulum which increases calcium release, leading to the activation of the protease calpain. Piezo proteins are fairly novel proteins, and whose sequences are not akin to other ion channels or proteins.
