Background: Podoplanin

Podoplanin (PDPN, Gp38 or Ots8) belongs to family of type-1 transmembrane sialomucin-like glycoproteins and is expressed in various tissues such as keratinocytes, synovial fibroblasts, lymphatic endothelial cells and cancerous cells. Originally identified in glomerular podocytes, PDPN is now considered as one of the most reliable markers for immunodetection of lymphatic endothelium. Its expression has been documented in benign/malignant cancers such as thymoma, osteosarcoma, laryngeal squamous cell carcinoma, odontogenic tumors as well as in CIS (carcinoma in situ)/ SCC (squamous cell carcinoma) of the oral mucosa. PDPN implicates in cellular migration and/or actin cytoskeleton organization, induction of platelet aggregation and is essential to lung cell proliferation as well as alveolus formation postpartum. PDPN expression is up-regulated by pro-inflammatory factors, such as IFN-gamma, TGF-beta, TNF-alpha and IL-1beta; and its mucin-type character has been shown to be associated with its pathophysiological functions including polarized migration of lymphatic endothelial cells as well as in invasion of cancer. PDPN knockout mice show embryonic lethality with respiratory failure and defective development of lymphoid follicles, germinal centers in spleen and lymph node remnants.
