Q: We purchased Protein Disulfide Isomerase/P4HB Antibody [NBP1-84051] and are wondering if it's specific for the monomeric version of the protein or the tetrameric version? We believe this antibody is staining primary cells (isolated from mice) that are not fibroblasts. Do you know if this antibody can stain primary cells that are not fibroblasts?
A: Our lab has not tested this Ab with the monomeric or multimeric subunits of the protein targets. However, the immunogen for NBP1-84051 is a.a. 236 - 379 of mouse protein (Uniprot/P09103) and since it is a polyclonal antibody (multiple epitopes), there are very strong chances that it could detect multimers also. Our lab has performed ICC/IF staining with NBP1-84051 on human U-2 OS cell line and we have not tested murine primary cells or fibroblasts yet. However, we strongly believe thatthis antibody will detect P4HB in any tissues/organs, cell lines/primary cells and fibroblasts as long as they express this protein.
