For each experiment, 500ul of DDK tagged RALBP1 overexpression lysates (at 1:5 dilution with HEK293T lysate), 2ug of anti-RALBP1 antibody and 20ul (0.1mg) of goat anti-mouse conjugated magnetic beads were mixed and incubated overnight. After extensive wash to remove any non-specific binding, the immuno-precipitated products were analyzed with rabbit anti-DDK polyclonal antibody.

Background: RALBP1

Can activate specifically hydrolysis of GTP bound to RAC1 and CDC42, but not RALA. Mediates ATP-dependent transport of S-(2,4-dinitrophenyl)-glutathione (DNP-SG) and doxorubicin (DOX) and is the major ATP-dependent transporter of glutathione conjugates of electrophiles (GS-E) and DOX in erythrocytes. Can catalyze transport of glutathione conjugates and xenobiotics, and may contribute to the multidrug resistance phenomenon. Serves as a scaffold protein that brings together proteins forming an endocytotic complex during interphase and also with CDC2 to switch off endocytosis, One of its substrates would be EPN1/Epsin

  • Q: Our customer would like to know if NBP2-02293 will recognize phospho-RALBP1 or not.

    A: NBP2-02293 was raised against full length human recombinant protein of human RALBP1. It was not raised against the phosphorylated protein and we cannot therefore guarantee that it will recognize the phosphorylated version. We have tested it against lysates however have not determined the phosphorylation status.
