Activity
Recombinant human Josephin-2 is a Ubiquitin-specific deconjugating enzyme.  Reaction conditions will need to be optimized for each specific application.  We recommend an initial Josephin-2 concentration of 20-100 nM when using Ubiquitin-rhodamine (U-555) substrate, and 100-500 nM when using Ubiquitin-AMC (U-550) substrate.

Carrier Free

What does CF mean?

CF stands for Carrier Free (CF). We typically add Bovine Serum Albumin (BSA) as a carrier protein to our recombinant proteins. Adding a carrier protein enhances protein stability, increases shelf-life, and allows the recombinant protein to be stored at a more dilute concentration. The carrier free version does not contain BSA.

What formulation is right for me?

In general, we advise purchasing the recombinant protein with BSA for use in cell or tissue culture, or as an ELISA standard. In contrast, the carrier free protein is recommended for applications, in which the presence of BSA could interfere.

Background: Josephin-2/JOSD2

Josephin-2 (also known as JOSD2) is a member of the Machado-Joseph Disease (MJD) family of cysteine proteases, whose other members include Ataxin-3, Ataxin-3-like and Josephin-1.  188 amino acids long with a predicted molecular weight of 21 kDa, JOSD2 is capable of cleaving K11- and to a lesser extent, K63-linked polyubiquitin chains in vitro.  This recombinant enzyme contains a c-terminal 6-His tag.
