Western Blot: SAV1 Antibody (3B2) [H00060485-M02] - MST2 & SAV1 increase the levels of PPAR gamma by increasing its protein stability.(A) Increasing amounts (0.5, 1, 3 µg) of Myc-SAV1 were co-expressed with Flag-PPAR gamma and/or HA-MST2 in 293 cells. At 48 h after transfection, cell lysates were immunoblotted with the indicated antibodies. (B) Flag-tagged PPAR gamma or PPAR gamma (128–505) was co-expressed with HA-MST2, HA-MST2-KD (inactive mutant), Myc-SAV1, and/or Myc-SAV1(1–201) in 293 cells as indicated. Cell lysates were prepared at various times after treatment with 40 µg/mL cycloheximide & then immunoblotted with an anti-Flag antibody. The PPAR gamma protein expression was quantified & the half-life of PPAR gamma protein was calculated. Expression of GAPDH was detected as a loading control. Background: SAV1

WW domain-containing proteins are found in all eukaryotes and play an important role in the regulation of a wide variety of cellular functions such as protein degradation, transcription, and RNA splicing. This gene encodes a protein which contains 2 WW domains and a coiled-coil region. It is ubiquitously expressed in adult tissues. The encoded protein is 94% identical to the mouse protein at the amino acid level.
