This affinity purified Slit2 Antibody was prepared from whole rabbit serum produced by repeated immunizations with a synthetic peptide corresponding aa 475-500 of Human Slit2 protein. (Uniprot: O94813)

Reactivity Notes

A BLAST analysis was used to suggest reactivity with this protein from human and chicken based on 100% homology for the immunogen sequence. Expect cross reactivity with SLIT-2 from Tetraodon nigroviridis, as only a single amino acid residue change is found within the immunogen sequence (94% positive by BLAST). The core 13 amino acids of the immunogen are also 100% homologous to SLIT-2 from mouse, rat, bovine and Xenopus laevis. Cross reactivity with SLIT-2 homologues from other sources has not been determined.

Western Blot: Slit2 Antibody [NB600-881] - Analysis of a band at 165 kDa (lane 1) corresponding to SLIT-2 present in a chicken spinal cord whole cell lysate (arrowhead). Approximately 30 ug of lysate was separated on a 4-20% Tris-Glycine gel by SDS-PAGE and transferred onto nitrocellulose. After blocking the membrane was probed with the primary antibody diluted to 1:1,350. Reaction occurred overnight at 4 C followed by washes and reaction with a 1:10,000 dilution of IRDye800 conjugated Gt-a-Rabbit IgG [H&L] MX for 45 min at room temperature (800 nm channel, green).

This affinity purified antibody has been tested for use in ELISA, IHC, and by western blot. Specific conditions for reactivity should be optimized by the end user. Expect a band at ~ 165 kDa corresponding to full-length protein by western blotting in the appropriate cell lysate or extract. For western blotting block the membrane with goat serum. Do not block with non-fat dry milk.

Background: Slit2

Slit proteins are ligands at the Roundabout (Robo) receptors and act as guidance cues in axonal migration/navigation during neural development, at the ventral midline of the neural tube. Slit1 and Slit2 are essential for midline guidance in the forebrain by acting as repulsive signals preventing inappropriate midline crossing by axons projecting from the olfactory bulb. A number of cleavage products are reported in the literature for Slit2 protiens (following alternate splicing). The C-terminal cleavage proteins are more diffusible than the larger N-terminal protein that is more tightly cell associated. Slit2 protein is expressed in fetal lung and kidney, and adult spinal cord. Weak expression in adult adrenal gland, thyroid, trachea.
