NPHP3-promoter plasmids were prepared from wildtype (wt.) promoter. (b–e) HeLa cells were transfected with pCMV-2B or pCMV-T beta 4 for 24 h. (b) HeLa cells were co-transfected with wildtype or mutant pEZX-PG02-NPHP3-promoter Gluc plasmid. & Gluc activity in cultured media was measured with luminometer using Gluc substrate. Bar graph indicates the mean of NPHP3-promoter activity. (c) Expression level of T beta 4 & NPHP3 transcripts were measured by RT-PCR. The cells were fixed & stained with antibody against Ac-tubulin (green) or NPHP3 (red). The representative fluorescence image of primary cilia was shown. (b) Overlay of fluorescence intensity of Ac-tubulin (green) & NPHP3 (red) through the whole length of primary cilia was shown in line graph (Line scan * → ** in a, right). (c,d) Cells were transfected with AccuTarget™ negative control siRNA (NC) or T beta 4-siRNA for 24 h. (c) The mRNA (upper) & protein (lower) expression of T beta 4 were shown. (d) The cells were incubated in serum-starved media for 36 h, fixed & stained with antibody against Ac-tubulin (green) & DAPI (blue). The ciliated cells in AccuTarget™ negative control siRNA-treated (white) & T beta 4-knockdown cells (grey) were counted (n > 500 cells).
