Immunocytochemistry/Immunofluorescence: Transgelin/TAGLN/SM22 alpha Antibody (SAIC-33B-129) - Azide and BSA Free [NBP3-20101] - Immunofluorescence analysis of paraformaldehyde fixed HeLa cells permeabilized with 0.15% Triton stained with the chimeric rabbit IgG version of SAIC-33B-129 (NBP3-20101) (1:100 dilution) for 1h followed by Alexa Fluor(R) 488 secondary antibody (1:1000 dilution), showing nuclear and cytoplasmic staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP3-20101, DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody followed by staining with Alexa Fluor(R) 488 secondary antibody.

Western Blot: Transgelin/TAGLN/SM22 alpha Antibody (SAIC-33B-129) - Azide and BSA Free [NBP3-20101] - HeLa(A), Caco-2(B), U2OS(C) cell lysate and human placenta(D), muscle(E) tissue lysates (35ug protein in RIPA buffer) were resolved on a SDS PAGE gel and blots were probed with the chimeric rabbit version of SAIC-33B-129 (NBP3-20101) at 0.0003 ug/ml before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.

Flow Cytometry: Transgelin/TAGLN/SM22 alpha Antibody (SAIC-33B-129) - Azide and BSA Free [NBP3-20101] - Paraformaldehyde fixed HeLa cells permeabilized with 0.5% Triton were stained with anti-unknown specificity antibody (isotype control, black line) or the rabbit IgG version of SAIC-33B-129 (NBP3-20101, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor(R) 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.

Background: Transgelin/TAGLN

In contrast to fast and slow skeletal muscle cells that fuse and terminally differentiate, smooth muscle cells are able to simultaneously proliferate and express lineage-restricted proteins. One of these proteins, expressed exclusively in smooth muscles, has been referred to as SM22-alpha, a 22 kDa protein with structural similarity to the vertebrate thin filament myofibrillar regulatory protein calponin and the Drosophila muscle protein mp20, neither of which play a direct role in the contractile apparatus.The protein is a transformation and shape-change sensitive actin cross-linking/geling protein found in fibroblasts and smooth muscle. Its expression is down-regulated in many cell lines, and this down-regulation may be an early and sensitive marker for the onset of transformation. A functional role of this protein is unclear.
