Western Blot: UbcH10/UBE2C Antibody (SAIC-41A-1) - Azide and BSA Free [NBP3-20107] - Caco-2(A) (0.3ug/ml), MCF7(B) (1ug/ml) and HeLa(C) (1ug/ml) cell lysates (35ug protein in RIPA buffer) were resolved on a SDS PAGE gel and blots were probed with the chimeric rabbit version of SAIC-41A-1 (NBP3-20107) before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.

Flow Cytometry: UbcH10/UBE2C Antibody (SAIC-41A-1) - Azide and BSA Free [NBP3-20107] - Paraformaldehyde fixed MCF7 cells permeabilized with 0.5% Triton were stained with anti-unknown specificity antibody (isotype control, black line) or the rabbit IgG version of SAIC-41A-1 (NBP3-20107, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor(R) 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.

Immunocytochemistry/Immunofluorescence: UbcH10/UBE2C Antibody (SAIC-41A-1) - Azide and BSA Free [NBP3-20107] - Immunofluorescence analysis of paraformaldehyde fixed Caco-2 cells permeabilized with 0.15% Triton stained with the chimeric rabbit IgG version of SAIC-41A-1 (NBP3-20107) (1:100 dilution) for 1h followed by Alexa Fluor(R) 488 secondary antibody (1:1000 dilution), showing NUCLEAR staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP3-20107, DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody followed by staining with Alexa Fluor(R) 488 secondary antibody.

Background: UbcH10/UBE2C

The modification of proteins with ubiquitin is an important cellular mechanism for targeting abnormal or short-lived proteins for degradation. Ubiquitination involves at least three classes of enzymes: ubiquitin-activating enzymes, or E1s, ubiquitin-conjugating enzymes, or E2s, and ubiquitin-protein ligases, or E3s. This gene encodes a member of the E2 ubiquitin-conjugating enzyme family. This enzyme is required for the destruction of mitotic cyclins and for cell cycle progression. Multiple alternatively spliced transcript variants have been found for this gene, but the full-length nature of some variants has not been defined.
