Detection of FXR/NR1H4 by Western Blot

Detection of FXR/NR1H4 by Western Blot

Generation of CRISPR knockout FXR mice. (A) Single cell embryos were injected with guide RNA that targets exon 5 of Fxr. The embryos were implanted into pseudo-pregnant females to generate founders. The sequencing results show that the mutation was a 47 base-pair deletion at exon 5. (B) Hepatocytes were isolated from control and Fxrex5 delta livers using the classic collagenase-perfusion method. Western blot was performed on whole cell lysates from control and Fxrex5 delta hepatocytes. VINCULIN was used as a loading control. (C) RNA-seq was performed on control and Fxr ex5 delta hepatocytes. Heatmap of 789 differentially expressed genes between control and Fxrex5 delta hepatocytes (p value and FDR value cut-off of 0.05). (D) Overlap of dysregulated genes from Fxrex5 delta hepatocyte RNAseq with Fxrex5 delta liver RNAseq obtained from Annak et al. (2011) [22]. Pathways analysis of all upregulated and downregulated genes was conducted using the KEGG and REACTOME databases. The cut-off for pathways analysis was chosen as p-value < 0.05, FDR < 0.1. The dotted line represents a FDR value of 0.05. Any value on the right of the dotted line signifies pathways that pass the FDR threshold of 0.05. (E) Hepatic and serum bile acid analysis at 4-week, 4-month and 12-month time-points in control and Fxrex5 delta animals (n = 5). (F) Expression analysis of selected downregulated and upregulated genes upon FXR deletion. The normalized FPKM values from RNAseq analysis were averaged for control and Fxrex5 delta hepatocytes and fold-changes were calculated (n = 3). T-test was performed to determine statistical significance relative to controls: * p < 0.05, ** p < 0.01. Image collected and cropped by CiteAb from the following open publication (https://pubmed.ncbi.nlm.nih.gov/36551676), licensed under a CC-BY license. Not internally tested by R&D Systems.

Background: FXR/NR1H4

Farnesoid X-activated Receptor (FXR, HRR-1, BAR, RIP14; NR1H4) is a member of the Orphan Nuclear Receptor family. FXR is expressed in the liver, intestinal villi, renal tubes and adrenal cortex. FXR is a global regulator of bile acid metabolism. Two genes, cholesterol 7a-hydroxylase (CYP7A1) and IBABP (Ileal Bile Acid Binding Protein), which are implicated in bile acid biosynthesis and recycling, respectively, are target genes of FXR. FXR is shown to be transcriptionally activated by farnesol metabolites such as farnesol itself and juvenile hormone III. FXR binds to DNA only as a heterodimer with Retinoid X Receptor (RXR).
