Immunocytochemistry/Immunofluorescence: B7-1/CD80 Antibody (16C10) - Chimeric [NBP3-12027] - Immunofluorescence analysis of paraformaldehyde fixed Daudi cells on Shi-fix(TM) coverslips stained with the chimeric rabbit IgG version of 16C10 (NBP3-12027) at 10 ug/ml for 1h followed by Alexa Fluor(R) 488 secondary antibody (2 ug/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP3-12027, DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody followed by staining with Alexa Fluor(R) 488 secondary antibody.

Flow Cytometry: B7-1/CD80 Antibody (16C10) - Chimeric - Azide and BSA Free [NBP3-12027] - Daudi cells were fixed using 2% PFA and stained with anti-unknown specificity antibody (isotype control, black line) or NBP3-12027 (blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.

Background: B7-1/CD80

The B-lymphocyte activation antigen B7-1 (CD80) provides regulatory signals for T lymphocytes as a consequence of binding to the CD28 and CTLA4 ligands of T cells. After engagement of T-cell receptor with antigen in association with major histocompatibility complex class II, a second signal mediated through the binding of B7 to CD28 greatly upregulates the production of multiple lymphokines (1). The relatively small CTLA-4/B7-1 binding interface exhibits an unusually high degree of shape complementarity. CTLA-4 forms homodimers through a newly defined interface of highly conserved residues. This zipper-like oligomerization provides the structural basis for forming unusually stable signalling complexes at the T-cell surface, underscoring the importance of potent inhibitory signalling in human immune responses (2).
