Background: CD44

CD44 is a type 1 transmembrane glycoprotein also known as Phagocytic Glycoprotein 1 (pgp 1) and HCAM. CD44 is the receptor for hyaluronate and exists as a large number of different isoforms due to alternative RNA splicing. The major isoform expressed on lymphocytes, myeloid cells, and erythrocytes is a glycosylated type 1 transmembrane protein. Other isoforms contain glycosaminoglycans and are expressed on hematopoietic and non hematopoietic cells. CD44 is involved in adhesion of leukocytes to endothelial cells, stromal cells, and the extracellular matrix.

  • Q: We used the CD44 variant 6 antibody (9A4, NB100-64818) in our Lab. I did not find the molecular weight of this antibody on the data sheet or in the related literature. I would appreciate it if you could give me some information about the molecular weight of this antibody.

    A: The lab has informed me that on testing of the immunogen the protein is expressed at 80-85 kDa, but when tested on endogenous samples the protein is picked up at approx. 200 kDa. Most likely the protein forms a dimer, or complex when picked up in endogenous samples. Additionally, I found a paper, PMC1853452 (https://www.ncbi.nlm.nih.gov/pmc/articles/PMC1853452/), that shows CD44v6 should be expressed at approx. 200 kDa - 250 kDa.
