Detection of CDK2 in HEK293, HeLa, K562 and Mouse Embryonic Fibroblast lysates by Western Blot.
Western blot shows lysates of HEK293 human embryonic kidney cell line, HeLa human cervical epithelial carcinoma cell line, K562 human chronic myelogenous leukemia cell line, and Balb/3T3 mouse embryonic fibroblast cell line. PVDF membrane was probed with 1 µg/mL Goat Anti-Human/Mouse CDK2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4654) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (HAF017). For additional reference, recombinant human CDK2, CDK4, and CDK6 (5 ng/lane) were included. A specific band for CDK2 was detected at approximately 34 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.Detection of p27/Kip1 in NRK, U2O2 and Mouse Embryonic Fibroblast lysates by Western Blot.
Western blot shows lysates of NRK rat normal kidney cell line, U2OS human osteosarcoma cell line, and Balb/3T3 mouse embryonic fibroblast cell line. PVDF membrane was probed with 0.2 µg/mL of Mouse Anti-Human/Mouse/Rat p27/Kip1 Monoclonal Antibody (Catalog # MAB2256) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for p27/Kip1 at approximately 30 kDa (as indicated). Western blot shows lysates of 1) mouse C2C12 and 2) mouse NIH 3T3 lysate. Cyclin D3 was detected in immersion fixed paraffin-embedded sections of human breast cancer tissue using Human Cyclin D3 Monoclonal Antibody (Catalog # MAB6570) at 15 µg/mL overnight at 4 °C. Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using the Anti-Mouse HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS002) and counterstained with hematoxylin (blue). Western blot shows lysates of MCF-7 human breast cancer cell line untreated (-) or treated (+) with 1 µM camptothecin (CPT) for 16 hours. PVDF membrane was probed with 0.5 µg/mL of Goat Anti-Human p21/CIP1/CDKN1A Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1047), followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017). A specific band was detected for p21/CIP1/CDKN1A at approximately 21 kDa (as indicated). CDK4 was detected in immersion fixed HeLa human cervical epithelial carcinoma cell line using Goat Anti-Human/Mouse CDK4 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5254) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (red; NL001) and counterstained with DAPI (blue). Specific staining was localized to cytoplasm and nuclei.
