Specificity

This monoclonal antibody recognizes the 56.5kDa (CK10); 50kDa (CK14); 50kDa (CK15); 48kDa (CK16); 40kDa (CK19) keratins of the acidic (Type I or LMW) subfamily. Twenty human keratins are resolved with two-dimensional gel electrophoresis into acidic (pI, 48, 46, 45, and 40kDa. monoclonal antibody AE3 recognizes the 65-67, 64, 59, 58, 56, and 52kDa keratins of basic subfamily. Many studies have shown the usefulness of keratins as markers in cancer research and tumor diagnosis. AE1/AE3 is a broad spectrum anti pan-keratin antibody cocktail, which differentiates epithelial tumors from non-epithelial tumors e.g. squamous vs. adenocarcinoma of the lung, liver carcinoma, breast cancer, and esophageal cancer.

Application Notes
Immunohistochemistry (Formalin-fixed): 1-2ug/ml for 30 min at RT. Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95C followed by cooling at RT for 20 minutes.
Optimal dilution for a specific application should be determined.

Background: Cytokeratin, LMW

This cytokeratin antibody recognizes most of the acidic (Type I) keratins. seen by western blot as bands between 40 and 56.5 kD. In keratinized epidermis 50 kD keratin is present in the basal layer, while 56.5 kD keratin is present in suprabasal layers. The cytokeratin AE1 antibody can be used to stain distinct cell layers in epithelial tissue and various neoplasms of epithelial origin.

  • Q: Can you tell me what the antigen sequence is for product number NBP2-29411 I guess most importantly I need to know if there is any cross-reactivity with mouse Krt77.

    A: NBP2-29411 was immunized against the full length protein. Since we don't epitope map our products, I will not have the specific homology to the mouse sequence, but I can provide you the overall homology. Overall homology is 64%. I would predict that there is a chance that this antibody will recognize the mouse protein. As we have not yet tested this product against mouse samples, I will not be able to say for certain.
