Scientific Data Images for Human LAP (TGF-beta 1) Antibody
LAP (TGF-beta 1) in HEK293 Human Cell Line.
LAP (TGF-beta 1) was detected in immersion fixed HEK293 human embryonic kidney cell line using Mouse Anti-Human LAP (TGF-beta 1) Monoclonal Antibody (Catalog # MAB246) at 3 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 493-conjugated Anti-Mouse IgG Secondary Antibody (green; Catalog # NL009) and counterstained with DAPI (blue). Specific staining was localized to cytoplasm. Recombinant Human LAP TGF-beta 1 (Catalog # 246-LP) inhibits Recombinant Human TGF-beta 1 (Catalog # 240-B) growth inhibition activity in the HT-2 mouse T cell line in a dose-dependent manner (orange line). Inhibition of Recombinant Human TGF-beta 1 (1 ng/mL) activity elicited by Recombinant Human LAP TGF-beta 1 (500 ng/mL) is neutralized (green line) by increasing concentrations of Human LAP TGF-beta 1 Monoclonal Antibody (Catalog # MAB246). The ND50 is typically 1-5 µg/mL.Detection of LAP (TGF-beta 1) by Western Blot
Specificity of R58 and L59 antibodies. (a) Schematic structure of a GST- rhLTGF-beta 1 protein generated in bacteria and a commercial rhLAP beta 1 generated in insect cells. The PLK cleavage site is indicated by arrowhead. (b-e) GST-LTGF-beta 1/rhLAP beta 1was incubated with/without PLK or PLN for 45 min at 37°C, and thereafter equal amounts of samples containing either 50 ng GST-rhLTGF-beta 1 or 500 ng rhLAP beta 1 were subjected to each lane in SDS-PAGE under reducing conditions followed by Western blot analyses using R58/L59 antibodies (b, d, respectively). Membranes used in panels b and d were reprobed with monoclonal (c) or polyclonal (e) anti-LAP antibodies. Lanes 1, intact GST-rhLTGF-beta 1 or rhLAP-beta 1; lanes 2, PLK digested GST-rhLTGF-beta 1 or rhLAP-beta 1; lanes 3, PLK; lane 4, PLN digested GST-rhLTGF-beta 1 or rhLAP-beta 1; lane 5, PLN. Representative results from three independent experiments with a similar result are shown. Application
Recommended Usage
Immunocytochemistry
3-25 µg/mL
Sample: Immersion fixed HEK293 human embryonic kidney cell line
Sample: Immersion fixed HEK293 human embryonic kidney cell line
Neutralization
Measured by its ability to neutralize LAP TGF‑ beta 1 inhibition of TGF‑ beta 1 growth inhibition in the HT‑2 mouse T cell line. Tsang, M. et al. (1995) Cytokine 7:389. The Neutralization Dose (ND50) is typically 1-5 µg/mL in the presence of 500 ng/mL Recombinant Human LAP TGF‑ beta 1 and 1 ng/mL Recombinant Human TGF‑ beta 1.
