Detection of Human PKC theta by Western Blot.

Western blot shows lysates of Jurkat human acute T cell leukemia cell line. PVDF membrane was probed with 1 µg/mL of Human/Mouse PKC theta Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4368) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for PKC theta at approximately 82 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Detection of Human PKC theta by Simple Western^TM.

Simple Western lane view shows lysates of Jurkat human acute T cell leukemia cell line, loaded at 0.2 mg/mL. A specific band was detected for PKC theta at approximately 82 kDa (as indicated) using 50 µg/mL of Sheep Anti-Human/Mouse PKC theta Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4368) followed by 1:50 dilution of HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

PKC theta (PRKCQ) is a calcium-independent phospholipid-dependent serine and threonine kinase that belongs to the novel PKC subfamily. PKC theta plays a critical role in several aspects of T cell biology. PKC theta knock out mice develop autoimmune disease. PKC theta functions through the activation of NFkB and AP1 transcription factors.
