Detection of Human/Mouse/Rat PPP2R4 by Western Blot. Western blot shows lysates of K562 human chronic myelogenous leukemia cell line, Raji human Burkitt's lymphoma cell line, CH‑1 mouse B cell lymphoma cell line, BaF3 mouse pro‑B cell line, and NRK rat normal kidney cell line. PVDF membrane was probed with 0.5 µg/mL of Human/Mouse/Rat PPP2R4 Monoclonal Antibody (Catalog # MAB5929) followed by HRP-conjugated Anti‑Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for PPP2R4 at approximately 39 to 41 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

PPP2R4, also known as serine/threonine protein phosphatase 2A regulatory subunit B’, PR53 isoform, and PTPA, is a widely expressed member of the PTPA-type peptidyl prolyl isomerase family. Human PPP2R4 is 358 amino acids (aa) in length. Three splicing variants produce four isoforms for PPP2R4, and isoform 2 is the standard protein. Isoform 1, isoform 3 and isoform 4 have deletions corresponding to aa 73-107, 45-108, and 73-149 in isoform 2, respectively. Human PPP2R4 is 87% aa identical to mouse and rabbit PPP2R4. PPP2R4 reversibly stimulates the variable phosphotyrosyl phosphatase activity of the PP2A core heterodimer in the presence of ATP and Mg2+. It has been shown that p53 downregulates the expression of PPP2R4.
