Detection of Human and Mouse Wnt‑10a by Western Blot.

Western blot shows lysates of Raji human Burkitt's lymphoma cell line and NIH-3T3 mouse embryonic fibroblast cell line. PVDF membrane was probed with 2 µg/mL of Rat Anti-Mouse Wnt-10a Monoclonal Antibody (Catalog # MAB34691) followed by HRP-conjugated Anti-Rat IgG Secondary Antibody (Catalog # HAF005). A specific band was detected for Wnt-10a at approximately 46 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: Wnt-10a

Wnt-10a is a presumably secreted, glycosylated member of the Wnt family of developmental proteins. Mouse Wnt-10a is synthesized as a 417 amino acid (aa) precursor that contains a 382 aa mature region. The mature region contains 24 cysteines and two potential N-linked glycosylation sites. Mature mouse Wnt-10a shares 96% and 99% aa sequence identity with mature human and rat Wnt-10a, respectively. Wnt-10a shares 64% aa sequence identity with Wnt-10b/12. Wnt-10a is produced by tumors and thymic epithelium.
