NF kappa B inhibitor alpha (IkB‑ alpha ) inhibits the NF kappa B complex by binding and sequestering it in the cytoplasm. Upon stimulation, IkB‑ alpha is phosphorylated on serine residues 32 and 36 marking it for degradation by the ubiquitin pathway. Following IkB‑ alpha degradation, the NF kappa B complex is able to translocate to the nucleus and activate transcription.

Detection of Human Phospho-IkB‑ alpha (S32/S36) by Western Blot.
Western blot shows lysates of HeLa human cervical epithelial carcinoma cell line untreated (-) or treated (+) with 20 ng/mL Recombinant Human TNF-a (Catalog # 210-TA) for 5 minutes. PVDF membrane was probed with 1 µg/mL Rabbit Anti-Human Phospho-IkB-a (S32/S36) Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4809) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). A specific band for Phospho-IkB-a (S32/S36) was detected at approximately 44 kDa (as indicated). For additional reference, duplicate lysates were probed with 0.1 µg/mL Human IkB-a Monoclonal Antibody (lower panel, Catalog # MAB4299).
