Benefits of Isolating Adult Stem Cells from Epithelial Tissue

Adult stem cell populations are found in most tissues of the human body and provide a long-lived source of self-renewing cells for both tissue homeostasis under normal conditions as well as for regeneration during disease or injury. In the adult, tissue specific stem cells can be either unipotent or multipotent. Tissue-specific adult stem cells exhibit an epigenetic memory conferring tissue specificity and regional identity. Moreover, adult stem cells may hold the keys to understanding aging, disease progression, and the development and spread of cancer. The MimEX™ Tissue Model System allow for the expansion and differentiation of ground-state, adult stem cell populations. These cells create sustainable and accessible 3-dimensional tissues in vitro and provide a valuable tool for biomarker discovery, disease modeling, drug screening*, and developmental biology studies.

The MimEX™ Tissue Processing Kit contains reagents and supplies for the collection and dissociation of adult epithelial tissue samples for the isolation of adult epithelial stem cells. The quantity of each component is sufficient to dissociate 1 tissue sample for stem cell isolation, up to 3 cm in size. Adult epithelial stem cells isolated using the tissue processing kit maintain local tissue regional identity, are genomically stable, and also preserve diseased tissue state.

 

Kit Contents

This kit contains the following reagents to isolate epithelial stem cells from biopsy tissue:

  • 30 mL of MimEX™ Tissue Collection Media
  • 250 mL of MimEX™ Stem Cell Isolation Media
  • 10 mL of MimEX™ Dissociation Solution
  • 2 Scalpels

Optimal Density of Adult Stem Cells Following Tissue Biopsy Dissociation and Digestion. Adult human intestinal epithelial tissue was digested and dissociated using the protocol described in the MimEX™ Tissue Collection Kit. A complete dissociation should yield mostly clusters of 2-5 cells. Some single cells and few large cell clusters may also be observed.

Briefly, process biopsy tissue with reagents in the MimEX™ Tissue Processing Kit using the following procedure:

  • Collect biopsy tissue using MimEX™ Tissue Collection Media
  • Dissect epithelial tissue from the biopsy
  • Dissociate tissue in MimEX™ Dissociation Solution
  • Resuspend dissociated tissue in MimEX™ Isolation Media
  • Plate tissue on MimEX™ Irradiated Fibroblasts in MimEX™ Expansion Media
  • Monitor for adult epithelial cell colony outgrowth

  • 30 mL of MimEX™ Tissue Collection Media
  • 250 mL of MimEX™ Stem Cell Isolation Media

  • Centrifuge
  • Hemocytometer

Procedure Overview

The protocol below describes the procedure for the isolation and expansion of human adult epithelial stem cells from biopsy tissue.

Note: The detailed protocol must be read in its entirety before using this product.

Isolation of Human Adult Epithelial Stem Cells from Biopsy Tissue

Prepare a 6-well plate with MimEX™ Irradiated Fibroblasts 1-3 days before culturing adult stem cells. Follow protocol for Plating MimEX™ Irradiated Fibroblasts.

Protocol for Isolation of Human Adult Epithelial Step 1

Harvest biopsy tissue and store it directly in MimEX™ Tissue Collection Media.

Protocol for Isolation of Human Adult Epithelial Step 2

Place tissue biopsy in a 10 cm petri dish.

Wash tissue 1-2 times in 5 mL of MimEX™ Stem Cell Isolation Media.

Protocol for Isolation of Human Adult Epithelial Step 3

Locate the epithelial surface of the tissue (identified by a yellow/orange appearance and lack of blood vessels and fat tissue).

Protocol for Isolation of Human Adult Epithelial Step 4

Isolate epithelial tissue using scissors and forceps.

Remove excess stroma and adipose tissue with fine forceps and dissecting scissors.

Protocol for Isolation of Human Adult Epithelial Step 5

Transfer tissue into a new 10 cm petri dish and, using two scalpels, mince the tissue.

Protocol for Isolation of Human Adult Epithelial Step 6

Collect tissue in a 50 mL centrifuge tube containing 10 ml of MimEX™ Dissociation Solution.

Shake tissue on an orbital shaker at 37 °C with ~120 rotations/minute for 30 minutes.

Triturate the tissue 10-20 times with a 5 mL pipette.

Protocol for Isolation of Human Adult Epithelial Step 8

Add 20 mL of MimEX™ Stem Cell Isolation Media, mix gently, and centrifuge at 200 × g for 6 minutes at 2-8 °C.

Discard supernatant, resuspend cell pellet in MimEX™ Stem Cell Isolation Media, and centrifuge at 200 × g for 8 minutes at 2-8 °C. Repeat 5 times.

Protocol for Isolation of Human Adult Epithelial Step 9

Re-suspend tissue pellet in 2.5 mL of prewarmed MimEX™ Expansion Media.

Add suspension to one well of a 6-well plate precoated with MimEX™ Irradiated Fibroblasts.

Protocol for Isolation of Human Adult Epithelial Step 10

Distribute cells in the well using the MimEX™ Cell Plating Procedure. (Critical Step)

Incubate the plate in a 37 °C/7.5% CO2 incubator. (Critical Step) without disturbing the plate.
