Product Summary

The Quantikine® Mouse Progranulin Immunoassay is a 4.5 hour solid phase ELISA designed to measure mouse Progranulin in cell culture supernates, tissue culture supernates, serum, and plasma. It contains NS0-expressed recombinant mouse Progranulin and has been shown to accurately quantitate the recombinant factor. Results obtained using natural Progranulin showed linear curves that were parallel to the standard curves obtained using the Quantikine® kit standards. These results indicate that this kit can be used to determine relative mass values for natural mouse Progranulin.

The recovery of mouse Progranulin spiked to three levels throughout the range of the assay in various matrices was evaluated.

To assess the linearity of the assay, samples containing high concentrations of mouse Progranulin were serially diluted with calibrator diluent to produce samples with values within the dynamic range of the assay. Samples were diluted prior to assay.

Background: Progranulin/PGRN

Progranulin, also known as Acrogranin, PCDGF, and Epithelin/Granulin Precursor, is a widely expressed glycoprotein that can undergo proteolysis to generate numerous Granulin peptides. Progranulin plays a role in the regulation of mesothelial differentiation, macrophage development, sexual differentiation of the brain, cartilage proliferation, wound repair, and the development of rheumatoid arthritis and osteoarthritis. It is often upregulated in cancer and contributes to tumor progression. Progranulin upregulation or mutation is associated with several neurological disorders including frontotemporal dementia, Creutzfeldt-Jakob disease, Alzheimer’s disease, and motor neuron disease.
