Background: Nanog

Expression of NANOG is required for the maintenance of pluripotency in epiblast and embryonic stem (ES) cells as well as for the ability to maintain ES self-renewal independently of LIF/Stat3. The role of NANOG in embryonic development suggested that it might be useful in the creation of stem cells that might be useful in cell replacement therapies in the treatment of several degenerative diseases. Artificial stem cells, termed induced pluripotent stem (iPS) cells, can be created by expressing POU5F1 (also known as Oct-4), another germline-specific transcription factor, and the transcription factors Sox2, Klf4 and Lin28 along with c-Myc in mouse fibroblasts. More recently, experiments have demonstrated that iPS cells could be generated using expression plasmids expressing NANOG, Sox2, KlfF4 and c-Myc, eliminating the need for virus introduction, thereby addressing a safety concern for potential use of iPS cells in regenerative medicine.

  • Q: As part of an Endometriosis-related project in horses, I would like to ask some information with regard to this. I would like to know about its tested or predicted reactivity with horse.

    A:

    This antibody has not yet been tested with horse. This means that use of this antibody with horse would not be covered by our guarantee; however, you would be eligible for our Innovator's Reward Program. For an indication of the likely cross-reactivity (how well the antibody is likely to perform with horse), I have run UniProt Blast sequence comparison of the immunogen sequence versus horse proteins. For Nanog Antibody NBP1-77109, the exact immunogen sequence is not available, so I have had to use the whole of the human protein, database reference EAW88651, for the sequence analysis. The result was a 76% match to F6PXN9 (https://www.uniprot.org/uniprotkb/F6PXN9/entry) and a 75% match to F6PPY5 (https://www.uniprot.org/uniprotkb/F6PPY5/history). Unfortunately these proteins from Equus caballus (horse) are unreviewed.
