Immunocytochemistry/Immunofluorescence: Nestin Antibody (4D11) [NBP1-92717] - Mixed cultures of neonatal rat neurons and glia stained with NBP1-92717 (red), chicken antibody to Vimentin (NB300-223, green) and DNA (DAPI stain, blue). Astrocytes and neuronal stem cells stain strongly and specifically in a clearly filamentous fashion with the Nestin (4D11) antibody. The filamentous staining pattern is as expected as both Nestin and Vimentin are components of 10nm filaments. Note that some cells contain Nestin, but do not stain strongly for Vimentin and so appear red. Others contain Vimentin and not Nestin and so appear green - these are likely to be fibroblastic or endothelial cells. Some cells express both proteins and so appear yellowish.

Background: Nestin

Nestin (NES) is a class VI intermediate filament protein which is unable to self-assemble or form homodimers because of a short NH2-terminus and is essential for the development of brain, eye etc. as well as for processes such as survival, renewal and mitogen-stimulated proliferation of neural progenitor cells. It facilitates the disassembly of phosphorylated vimentin intermediate filaments (IF) during mitosis and the trafficking/distribution of IF proteins or other factors to daughter cells during progenitor cell division. Nestin is expressed in dividing cells during early stages of development e.g. CNS, PNS, myogenic or other tissues etc, whereas with differentiation, it is downregulated followed by replacement with tissue-specific intermediate filament proteins, and therefore, is widely used as a neuronal stem cell marker. Under pathological conditions, nestin is expressed in repair processes in CNS, muscle, liver, and infarcted myocardium, and in tumors, increased nestin correlates with aggressive growth, metastasis, and poor prognosis. Furthermore, nestin is more specifically expressed in proliferating small-sized tumor vessels in glioblastoma and gastric, colorectal, and prostate cancers than are other tumor vessel markers, indicating nestin's use as a marker for newly synthesized tumor vessels.
