Western analysis of cell lysates.a Western analysis of primary neurons, astrocytes, microglia, and OPCs derived from WT and PINK1 KO rats. Cultured cells were treated with 100 nM valinomycin or DMSO control for 4 h at 37 C, then harvested for western analysis. Ten micrograms of cell lysate protein was loaded in each lane, resolved by SDS-PAGE, transferred to PVDF membrane, and blotted using anti-pS65-Ub antibody. The same membrane was re-probed using the antibodies indicated on the lower panels as markers for neurons ( beta 3-tubulin), astrocytes (ALDH1L1), microglia (Iba-1), and OPCs (Olig-2). b Mean +/− SEM pS65-Ub level measured by densitometry of western blot membranes from four independent experiments. Background: Olig2

Olig2 is a transcription factor which is required for oligodendrocyte and motor neuron specification in the spinal cord, as well as for the development of somatic motor neurons in the hindbrain. Expressed in the ventral spinal cord as early as 9.5 dpc and scattered in the mantle zone, likely corresponding to oligodendrocyte progenitors migrating out from their site of origin. (Arnett et al., 2004) From 10.5 through 14.5 dpc, Olig2 is expressed in numerous cells in the ventricular and subventricular zones of the lateral and medial ganglionic eminences, suggesting that expression might not be limited to the oligodendrocytic lineage. Olig2 has further been shown to be critical in the proliferation of malignant glioma in brain tumors (Ligon et al., 2007).
