Background: PDE8B

Cyclic nucleotide phosphodiesterases (PDEs) catalyze the hydrolysis of the 5-prime,3-prime-cyclic nucleotides cAMP and cGMP to the corresponding nucleoside 5-prime-monophosphates. Mammalian PDEs have been grouped into several families based on substrate affinities, inhibitor sensitivities, mode of regulation, and amino acid sequence homologies. The PDE8 family contains high-affinity cAMP-specific, IBMX (3-isobutyl-1-methyl-xanthine)-insensitive PDEs. All PDEs share a conserved C-terminal catalytic region and a variable N-terminal domain that presumably accounts for the distinctive regulatory properties unique to the individual families. PDE8B exhibits cAMP PDE activity which was not inhibited by various PDE inhibitors, including IBMX. The cAMP hydrolytic activity was unaffected by cGMP, and no cGMP PDE hydrolysis has been detected. Northern blot analysis indicated that the 4.2-kb PDE8B mRNA was expressed at the highest levels in thyroid, and at lower levels in brain, spinal cord, and placenta.
