Protein Array: PU.1/Spi-1 Antibody (PU1/2146) [NBP2-75738] - Analysis of Protein Array containing more than 19,000 full-length human proteins using PU.1/Spi-1 Antibody (PU1/2146)Z- and S- Score: The Z-score represents the strength of a signal that a monoclonal antibody (MAb) (in combination with a fluorescently-tagged anti-IgG secondary antibody) produces when binding to a particular protein on the HuProt(TM) array. Z-scores are described in units of standard deviations (SD's) above the mean value of all signals generated on that array. If targets on HuProt(TM) are arranged in descending order of the Z-score, the S-score is the difference (also in units of SD's) between the Z-score. S-score therefore represents the relative target specificity of a MAb to its intended target. A MAb is considered to specific to its intended target, if the MAb has an S-score of at least 2.5.

Applications for PU.1/Spi-1 Antibody (PU1/2146)

Application
Recommended Usage

Flow Cytometry

1-2 ug/million cells

Immunocytochemistry/ Immunofluorescence

1-2 ug/ml

Immunohistochemistry

1-2 ug/ml

Immunohistochemistry-Paraffin

1-2 ug/ml

Western Blot

1-2 ug/ml
Application Notes
Immunohistochemistry (Formalin-fixed): 1-2ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95C followed by cooling at RT for 20 minutes.
Optimal dilution for a specific application should be determined.

Background: PU.1/Spi-1

PU.1/Spi1 encodes an ETS-domain transcription factor that activates gene expression during myeloid and B-lymphoid cell development. The nuclear protein binds to a purine-rich sequence known as the PU-box found near the promoters of target genes, and regulates their expression in coordination with other transcription factors and cofactors. The protein can also regulate alternative splicing of target genes. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq]
