Flow Cytometry: Rab7a Antibody (10E4) [NBP2-60237] - Analysis of RAB7A in HeLa cells. The cell was stained with at 2-5ug for 1x106cells (red). A Goat anti mouse IgG (Alexa fluor 488) was used as the secondary antibody. Mouse monoclonal IgG was used as the isotypecontrol (dark gray), cells without incubation with primary and secondary antibody was used as the negative control (light gray).

Western Blot Shows Rab7a Specificity Using Knockout Cell Line.

Western blot shows lysates of HeLa human cervical epithelial carcinoma parental cell line and Rab7a knockout HeLa cell line (KO). Nitrocellulose membrane was probed with Rab7a Antibody (10E4) - BSA Free (Catalog # NBP2-60237) followed by HRP-conjugated secondary antibody. A specific band was detected for Rab7a at approximately 23.4 kDa (as indicated) in the parental HeLa cell line, but is not detectable in knockout HeLa cell line. Primary antibody dilution used: 1/1000. The Ponceau stained transfer of the blot is shown. This experiment was conducted under reducing conditions. Background: Rab7a

RAB7 is a RAB family member that regulates vesicle traffic in the late endosomes and also from late endosomes to lysosomes. This encoded protein is also involved in the cellular vacuolation of the VacA cytotoxin of Helicobacter pylori. Mutations at highly conserved amino acid residues in this gene have caused some forms of Charcot-Marie-Tooth (CMT) type 2 neuropathies.
