  Human A-172 glioblastoma cells stained with early endosomal marker Rab7a (NBP1-87174).

  Cell were grown on coverslips overnight, media was aspirated and cells fixed with 4% paraformaldehyde for 20 min. Following 3x washes with PBS, cells were permeabilized with ice-cold methanol for 10 min. Following 3x washes, cells were blocked overnight with blocking buffer (0.3% triton-X 100, 5% serum in PBS). The following day, cells were incubated with primary Rab7a antibody (1:200) for 1 h at RT. Following 3x washes in PBS, cells were incubated with anti-rabbit AF488 secondary antibody for 1 h at RT. Following incubation, cells were washes 3x with PBS, mounted on slides using Prolong Gold (with DAPI) and imaged using a Nikon Confocal microscope.
