Histone H1.2 Antibody (16H4)

Immunohistochemistry: Histone H1.2 Antibody (16H4) [NBP3-26232] -

Immunohistochemistry: Histone H1.2 Antibody (16H4) [NBP3-26232] - Image of Histone H1.2 Antibody (16H4) diluted at 1:50 and staining in paraffin-embedded human stomach tissue performed. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.32% DAB.

Immunocytochemistry/Immunofluorescence: Histone H1.2 Antibody (16H4) [NBP3-26232] -

Immunocytochemistry/Immunofluorescence: Histone H1.2 Antibody (16H4) [NBP3-26232] - Staining of Hela with Histone H1.2 Antibody (16H4) at 1:30, counter-stained with DAPI. The cells were fixed in 4% formaldehyde and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4C. The secondary antibody was Alexa Fluor 504-conjugated Goat Anti-Rabbit IgG (H+L).

Flow Cytometry: Histone H1.2 Antibody (16H4) [NBP3-26232] -

Flow Cytometry: Histone H1.2 Antibody (16H4) [NBP3-26232] - Overlay Peak curve showing Hela cells stained with Histone H1.2 Antibody (16H4) (red line) at 1:50. The cells were fixed in 4% formaldehyde and permeated by 0.2% Triton X-100. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*10^6 cells) for 45min at 4C. The secondary antibody used was FITC-conjugated Goat Anti-rabbit IgG (H+L) at 1:200 dilution for 35min at 4C. Control antibody (green line) was rabbit IgG (1ug/1*10^6 cells) used under the same conditions. Acquisition of >10,000 events was performed.

Background: Histone H1.2

Histones are nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber as they play a critical role in transcription regulation, DNA repair, DNA replication, and chromosomal stability. The Histone H1.2 protein is 213 amino acids long, 21 kDA and is encoded by the HIST1H1C gene. H1 proteins are essential in forming chromatin fiber and for the condensation of nucleosome chains into higher-order structured fibers. The HIST1H1C gene has been investigated in leukemia and parkinson's disease. It plays a role in apoptosis, PKA signaling, cell cycle chromosome condensation in prometaphase, and histone modification through interactions with various genes such as TONSL, NPM1, YWHAQ, EED, and ESR2.

Alternate Names
