Western blot shows Trastuzumab, Adalimumab (negative control), Brentuxumab (negative control), and Gemtuzumab (negative control) biosimilars. PVDF membrane was probed with 2 µg/mL of Anti- Trastuzumab (Anti-Idiotype) Monoclonal Antibody (Catalog # MAB9547) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for Trastuzumab at approximately 240 kDa (as indicated). This experiment was conducted under non-reducing conditions and using Immunoblot Buffer Group 1.

Biotinylated biosimilars were serially diluted and incubated with Anti-Trastuzumab (Anti-Idiotype) Antibody (Catalog # MAB9547, green line) coated at 1 μg/mL on a 96-well polystyrene plate. No cross-reactivity with Adalimumab (black line) or Gemtuzumab (orange line) was observed.

A panel of biosimilars was coated at 1 μg/mL in 96-well polystyrene plates. Anti-Trastuzumab (Anti-Idiotype) Antibody (Catalog # MAB9547, 333 ng/mL) detected Trastuzumab. No cross-reactivity with Cetuximab, Ibritumomab, Rituximab, Adalimumab, Tositumomab, Brentuximab, Gemtuzumab, Solanezumab, Teprotumumab, and Aducanumab was observed.

Use a manual defrost freezer and avoid repeated freeze-thaw cycles.

The anti-Trastuzumab antibody is an anti-Idiotype antibody raised against and selected for the antigenic determinants of the variable region of Trastuzumab. The antibody has been tested against a broad range of other biosimilars to ensure specificity. This highly specialized anti-idiotypic antibody can be used in a variety of pharmacokinetic (PK) assay formats.
