Protein Array: beta-Catenin Antibody (CTNNB1/2030R) [NBP2-79753] - Analysis of Protein Array containing more than 19,000 full-length human proteins using beta-Catenin Antibody (CTNNB1/2030R). Z- and S- Score: The Z-score represents the strength of a signal that a monoclonal antibody (MAb) (in combination with a fluorescently-tagged anti-IgG secondary antibody) produces when binding to a particular protein on the HuProt(TM) array. Z-scores are described in units of standard deviations (SD's) above the mean value of all signals generated on that array. If targets on HuProt(TM) are arranged in descending order of the Z-score, the S-score is the difference (also in units of SD's) between the Z-score. S-score therefore represents the relative target specificity of a MAb to its intended target. A MAb is considered to specific to its intended target, if the MAb has an S-score of at least 2.5.

Applications for beta-Catenin Antibody (CTNNB1/2030R)

Application
Recommended Usage

Flow Cytometry

1-2 ug/million cells

Immunocytochemistry/ Immunofluorescence

1-2 ug/ml

Immunohistochemistry

1-2 ug/ml

Immunohistochemistry-Paraffin

1-2 ug/ml

Western Blot

1-2 ug/ml
Application Notes
Immunohistochemistry (Formalin-fixed): 1-2ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95C followed by cooling at RT for 20 minutes.
Optimal dilution for a specific application should be determined.

Background: beta-Catenin

The protein encoded by the beta Catenin gene is part of a complex of proteins that constitute adherens junctions (AJs). AJs arenecessary for the creation and maintenance of epithelial cell layers by regulating cell growth and adhesion betweencells. The encoded protein also anchors the actin cytoskeleton and may be responsible for transmitting the contactinhibition signal that causes cells to stop dividing once the epithelial sheet is complete. Finally, this proteinbinds to the product of the APC gene, which is mutated in adenomatous polyposis of the colon. Mutations in this geneare a cause of colorectal cancer (CRC), pilomatrixoma (PTR), medulloblastoma (MDB), and ovarian cancer. Three transcript variants encoding the same protein have been found for this gene.
