Western Blot Shows DDX1 Specificity Using Knockout Cell Line.

Western blot shows lysates of U2OS human osteosarcoma parental cell line and DDX1 knockout U2OS cell line (KO). Nitrocellulose membrane was probed with DDX1 Antibody (3E5E2) - BSA Free (Catalog # NBP2-61745) followed by HRP-conjugated secondary antibody. A specific band was detected for DDX1 at approximately 82.4 kDa (as indicated) in the parental U2OS cell line, but is not detectable in knockout U2OS cell line. Primary antibody dilution used: 1/500. U2OS human osteosarcoma cell line lysates were prepared and immunoprecipitation was performed using 2.0 μg of DDX1 Antibody (3E5E2) - BSA Free (Catalog # NBP2-61745) pre-coupled to Dynabeads Protein G. Immunoprecipitated DDX1 was detected in Western Blot with a rabbit DDX1 antibody used at 1/1000. The Ponceau stained transfer of the blot is shown. Background: DDX1

DEAD box proteins, characterized by the conserved motif Asp-Glu-Ala-Asp (DEAD), are putative RNA helicases. They are implicated in a number of cellular processes involving alteration of RNA secondary structure such as translation initiation, nuclear and mitochondrial splicing, and ribosome and spliceosome assembly. Based on their distribution patterns, some members of this family are believed to be involved in embryogenesis, spermatogenesis, and cellular growth and division. This gene encodes a DEAD box protein of unknown function. It shows high transcription levels in 2 retinoblastoma cell lines and in tissues of neuroectodermal origin.;
