Western blotting analysis using FoxO1/FKHR antibody. FoxO1/FKHRexpression in wild type (WT) and FoxO1/FKHRshRNA knockdown (KD) HeLa cells with 30 ug of total cell lysates. GAPDH serves as a loading control. The blot was incubated with FoxO1/FKHR antibody(1:5,000) and HRP-conjugated goat anti rabbit secondary antibody (1:50,000) respectively. Image was developed using FeQ(TM) ECL Substrate Kit.

Background: FoxO1/FKHR

FoxO1, also called FKHR (forkhead in rhabdomyosarcoma), is a ubiquitously expressed member of the forkhead box O family of winged helix transcription factors. In neurons, it is activated in response to stress, translocating to the nucleus where it promotes apoptosis and blocks proliferation. In insulin-responsive tissues, nutrient abundance triggers phosphorylation by AKT that blocks nuclear translocation and activity.
