T98G cells were treated with LLOMe (1 uM) for 1 h to induce lysosomal damage. Cells were fixed (4% paraformaldehyde) 6 h after washout, blocked with 10% goat serum in 0.1% PBST (1 h, RT), stained with DyLight 488 conjugated galectin-3 antibody (green) (overnight, 4C).The following day, samples were washed with cold PBST, mounted using Prolong Gold mountant with DAPI (blue). Flow Cytometry: Galectin-3 Antibody (A3A12) [NB300-538] - Using the Alexa Fluor 647 direct conjugate An intracellular stain was performed on HT-29 cells with Galectin-3 (A3A12) antibody NB300-538AF647 (blue) and a matched isotype control NBP1-97005AF647 (orange). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 5 ug/mL for 30 minutes at room temperature.
